I need to develop web GIS application with shapefiles of roads, counties, cities (different zoom levels). I would use Map Suite WMS Server edition. User will be able to write city name in search and he will get city map with legend and data from mysql database. I want to ask you if I can run this application on my local server or do I need some VPS server? Also, I want to ask where I can find those shapefiles I need (for USA).

Thanks for your post, you may want to actually take a look at the Map Suite Web Edition as it will also meet your requirements and allow you to do the rendering and search, while the WMS Server edition is primarily built to provide highly scalable rendering of tiles. You can run your application on your own serve so that wont be a problem.
As for the shapefiles you can download them for several countries including the USA from our open street map server at:
